Karakterisasi Hasil Degradasi Zat Warna Bromocresol Purple Oleh Lakase Trichoderma Asperellum Lbkurcc1 Menggunakan Hplc, Ftir Dan Uji Toksisitas

Trichoderma asperellum LBKURCC1 is a biocontrol fungi capable of producing the enzyme laccase. Laccase (EC 1.10.3.2) is an extracellular enzyme that contains copper ions and catalyzes the four-electron oxidation of phenolic substrates. From previous studies, T. Asperellum LBKURCC1 laccase was proven to be able to oxidize several triphenylmethane dyes, including Bromocresol purple (BCP), as shown by changes in its visible light absorbtion spectra. As an initial step to determine the mechanism of BCP degradation by T. asperellum LBKURCC1 laccase, in this study compounds formed from the incubation of BCP with crude extracts of T. asperellum LBKURCC1 laccase were analyzed using High Performance Liquid Chromatography (HPLC). Changes in the chromatogram pattern were compared to that of pure BCP crystals. Compared to BCP control solution in buffer, BCP incubated with inactivated crude laccase showed several additional peaks representing degradation products after 6 days incubation. These additional peaks were already seen on day 0 in BCP incubated with active crude laccase. Although no additional peaks were found in BCP control solution in buffer after 6 days of incubation, the single peak belonging to BCP became slightly more polar. In addition, the FTIR pattern was compared to determine changes in functional groups that occurred in BCP dye incubated with crude extract of laccase T. asperellum LBKURCC1. The FTIR results did not show any addition or reduction of functional groups in the control BCP which was incubated with buffer, inactive enzymes and tested with active enzymes. BCP dye incubated with crude extract of laccase T. asperellum LBKURCC1 was able to transform from a sulfonic structure (yellow) to a cyclic sulfonate ester (purple) even in a buffer pH of 5,5, beginning day 4 of incubation. This transformation in the same buffer pH was not observed in control BCP incubated in buffer alone. This study also determined the toxicity of BCP incubated with laccase using the Brine Shrimp Lethality Test method. BCP had medium toxicity with an LC50 of 245 μg/mL. After the addition of in-activated and active crude laccase extracts to BCP, the toxicity was rapidly eliminated after only 30 minutes incubation, with LC50>1000. This shows the
