Isolasi Dan Karakterisasi Kolagen Sisik Ikan Gabus (Channa Striata) Hasil Budidaya Dan Tangkapan Di Alam
Snakehead fish (Channa striata) is one of the freshwater fish with high economic value and much favored by the public because it has a meaty texture and a savory taste. Snakehead fish are often found in nature such as swamp waters. But now snakehead fish have been widely cultivated with media in the form of tarpaulin ponds. Pond raised snakehead fish and wild sources has a habitat with different external factors. Currently, research about snakehead fish caught in the wild has been widely carried out, but for pond raised snakehead fish, it is still rarely done, especially regarding scale collagen from snakehead fish. Snakehead fish scales are parts of the body that are not consumed and are still rarely used optimally. This of course, can increase the economic value of scales which were initially only in the form of waste but can be isolated into collagen which has high economic value.
The aims of this study were to obtain scale collagen from snakehead fish that were pond raised and caught in the wild, to determined the characteristics of the scale collagen from snakehead fish that were pond raised and caught in the wild, and to analyze the differences in scale collagen from snakehead fish that were pond raised and wild sources. This research is expected to provide scientific information to the public or readers about the scale collagen of pond raised snakehead fish and wild sources.
The research was conducted in February to May 2022 at the Laboratorium Pengolahan Hasil Perikanan dan Laboratorium Kimia Hasil Perikanan Fakultas Perikanan dan Kelautan, Laboratorium Sains dan Mineral Universitas Riau, Balai Standarisasi dan Pelayanan Jasa Industri Pekanbaru, Laboratorium Jasa Pengujian dan Sertifikasi Institut Pertanian Bogor, Laboratorium Terpadu Universitas Diponegoro dan Laboratorium Scanning Electron Microscope Institut Teknik Bandung. The main ingredients in this study were scales from pond raised snakehead fish taken from one of the residents who cultivated snakehead fish in Kampung Sawit Permai, Kecamatan Dayun, Kabupaten Siak, Riau. Meanwhile, the scales of wild sources snakehead fish were obtained from the waters of Danau Zamrud, Kecamatan Dayun, Kabupaten Siak, Riau. The materials and tools used are all supporting chemical materials and tools and other supporting materials.
This study used an experimental method and its implementation was carried out in two interrelated stages, which included preparation of fish scales from pond raised and caught snakehead fish in the wild to produce scale flour and then proximate analysis and yield calculations were carried out. The second stage is the isolation of collagen which begins with the extraction process using a strong acid, namely HCl. The collagen obtained was characterized in the form of functional groups, peptide content, amino acid content and morphology. The data obtained were analyzed statistically according to the research stages using SPSS

28 software with the Independent Samples T Test design method to see if there was a difference between the two samples.
The scales from pond raised snakehead fish are slimy, brownish and pale, while the scales from fish wild sources are gray and shiny. Fish scales from pond raised snakehead contain higher levels of protein and fat than caught in the wild, but ash and carbohydrates are lower for pond raised fish than wild sources. The yield of scale collagen from pond raised snakehead fish was 15.82%, higher than the wild sources of 14.27%. Morphologically, the scale collagen from pond raised snakehead fish has a smooth, tight and dense surface. While the wild sources looks like a small lump on the surface. Scale collagen from pond raised snakehead fish is higher in protein and ash content but lower in fat and carbohydrates than wild sources. The amino acid composition of scale collagen from pond raised snakehead fish was higher in leucine and glycine but lower in lysine compared to wild catch. Functional group analysis showed that both scale collagen contained amide A, amide B, amide I, amide II, and amide III which were indicated as type 1 collagen. Collagen peptide of pond raised snakehead fish scales was Arg-Pro (RP) with a molecular weight of 271.316 Da, while the wild sources snakehead fish scale collagen peptide is Arg-Gly-Ser (RGS) with a molecular weight of 318.330 Da. Based on their characteristics, the two collagens isolated from snakehead fish scales can have promising applications in the food, medical, and cosmetic industries, where pond raised snakehead fish scale collagen is better than wild sources.
