Sea cucumbers are included in one of the marine biotas with high economic
value and export commodities with health benefits. Based on research that has been
done, sea cucumber H. Atra has antibacterial activity and toxic activity against
cancer cells. The ability of sea cucumbers in the health sector is related to the
metabolic compounds contained, one of which is saponins.
The purpose of this study was to determine the saponin compounds
contained in the kasur sea cucumber fraction, to test sea cucumber isolates as
anticancer, and to obtain the best concentration of saponin isolates against cancer
cells. The benefits resulting from this research, are that sea cucumbers have the
potential as anticancer, provide information on the importance of developing and
cultivating sea cucumbers, and provide opportunities for marine biota to develop
in the fishery product processing industry and the health industry.
The research was carried out from June 2022 to December 2022 at the
Chemical Laboratory (FAPERIKA-UNRI), Lab. Organic Chemistry (FMIPAUNRI),
Lab. Organic Chemistry (FMIPA-UNP), and Labs. Primate Animal Study
Centre (PSSP) Bogor Agricultural University. The main ingredient in this study
was the active fraction of sea cucumbers that had gone through the fractionation
process. The research method used is an experimental method with descriptive data
analysis and completely randomized design analysis (CRD). The research
procedure was carried out in 3 stages, stage 1 testing of saponin metabolic
compounds, stage 2 isolation of saponin compounds, and stage 3 toxicity tests on
cancer cells using the MTT assay.
The results of the secondary metabolite compound test showed that the
active fraction of sea cucumbers were positive for steroid saponins with the
formation of foam and a bluish green colour. Isolation of the active fraction
compound from the sea cucumber was analysed by thin layer chromatography, the
stain formed had a retention factor value of 0.9, and a blue colour appeared after
being given Liberman-Burchard (strong acid) observed through UV 254-366nm so
that it could be declared positive for containing saponin compounds. In the toxicity
test, the ethyl fraction was very active against HeLa cancer cells (5.82 g/ml) and
active (11,404 g/ml) MCF-7 cells. The hexane fraction was very active (8,132 g/ml)
HeLa cells and active (11,745 g/ml) MCF-7 cells as comparison doxorubicin was
very active (2,084 g/ml) HeLa cells and very active (1,587 g/ml) MCF-7 cells. The
best concentration of steroid saponin isolates had a very significant effect on HeLa
cells. The concentration of saponins with very active toxicity is 15.62 g/mL with an
inhibitory power of 12.10%.
