Analisis Sekuens Barkode Dna Pada Daerah Trnl-Trnl-Trnf Intergenic Spacer Tumbuhan Terong Asam Bulu (Solanum Ferox L.)
Sour eggplant belongs to the genus Solanum used in this study, which has villosus and not easy to fall out on the skin of the fruit that distinguishes it from other sour eggplant. DNA barcoding is a technique for identification using short DNA sequences from the genome of an organism. The objective of this research is to analyze the trnL-trnL-trnF intergenic spacer of the sour eggplant (S. ferox L.) which was taken in Rokan Hilir Regency, Province of Riau. The research procedure is morphological observations, and the molecular procedures are sample preparation, total DNA isolation using the Geneaid kit, total DNA electrophoresis, PCR using the DreamTaq DNA polymerase (Thermo Scientific) kit, electrophoresis of PCR products, sequencing, and data analysis using the BioEdit, BLASTn and MEGA versions. 6.0. The results of this study indicated that the DNA sequence in the trnL-trnL-trnF intergenic spacer region of the sour eggplant was 1.091 bp in size and there was no percent identity value that reaches 100% with the sequences available in Genbank. The highest identity value was found in S. vestissimum (98.81%) and the lowest with S. hirtum (97.71%).
