Seleksi Bakteri Rhizosfer Padi Ladang Penghasil Indole Acetic Acid (Iaa) Dan Identifikasi Berdasarkan Sequen Gen 16s Rrna
Rhizosphere bacteria are bacteria that live ini the root area (rhizosphere) and play an important role in plant growth. In general, bacteria in the plant rhizosphere provide various benefits to plants. One of them is production of growth hormone, Indole Acetic Acid (IAA). This study aims to determine the potential of eight exopolysaccharide-producing bacteria to produce IAA hormone ad identify the 16S rRNA gene sequences of potential bacterial isolates to see their diversity. Bacterial isolates were grown on Nutrient Broth + L-tryptophan 500 μg/ml media as a precursor for 3 days and tested qualitatively using Salkowski’s reagent. The result of this study was 4 isolates that changed color when reacted with Salkowski’s reagent. Quantitative test using a spectrophotometer at a wavelength of 530 nm shown the highest IAA obtained in EPS 1 and EPS 6, it’s 60,83±4.65 and 49.95±3.22 ppm. Based on identification of the 16S rRNA gene sequence, EPS 1 and EPS 6 were identical to Klebsiella variicola strain 13450 with 100% homology.
