Isolasi Dan Seleksi Dark Septate Endophyte Dari Tanaman Cabai, Terung, Dan Sawi Sebagai Agen Biokontrol Penyakit Pada Eukaliptus
Eucalyptus sp. tree has great potential to be developed as raw material for pulp and paper because it has fast-growing properties, good adaptability and high productivity. Monoculture crops with low vegetation diversity have the potential to cause disease epidemics caused by the pathogen Cylindrocladium sp. (leaf spot), Ceratocystis sp. (stem rot), and Ganoderma philippii (root rot). The integrated control has not been satisfactory, so other control alternatives are used using dark septate endophyte (DSE) fungi. This study aims to isolate and select DSE isolates from chilli, eggplant, and mustard greens as biocontrol agents for Cylindrocladium sp., Ceratocystis sp., and G. philippii which infects Eucalyptus sp. Observation of DSE colonization in roots was performed microscopically, and DSE isolation was performed using corn meal agar medium. The DSE isolates obtained were tested for their ability to inhibit several fungal pathogens in the laboratory. Biosafety tests in hemolysis and pathogenicity were carried out on the five best DSE isolates obtained using the Analytical Hierarchy Process (AHP) method. The results obtained 25 isolates of DSE fungi with the typical characteristics of septate and pigmented hyphae. These isolates were able to inhibit the development of pathogenic fungi with varying degrees of inhibition. The five best DSE isolates based on growth rate and their ability to inhibit fungal pathogens were T2.S8, T3.S3, T3.S1, T2.S6, and T4.S3. Blood agar and pathogenesis tests show that they are not pathogenic for humans, animals and plants. Based on the macroscopic and microscopic characteristics, the DSE isolate was identified as Curvularia sp. These DSE isolates have the potential to be developed as alternative antagonist agents.
