Aktivitas Ekstrak Kasar Rumput Laut Coklat (Sargassum Sp.) Sebagai Antioksidan Dan Inhibitor Alfa Glukosidase
ABSTRACT
Sargassum sp. is one of brown seaweed that contains bioactive compounds. The bioactive compounds in seaweed acts as a food and non-food (pharmaceutical) ingredient. This study was aimed to identify bioactive compounds (flavonoids, alkaloids, steroids/terpenoids, saponins, and phenols), total phenolics, antioxidant activity, and alpha-glucosidase inhibitors from the crude extract of Sargassum sp. This study used an experimental method by extracting Sargassum sp. with solvents of different polarity. The solvent was used consisted of n-hexane, ethyl acetate, and methanol. The analysis parameters consisted of yield, phytochemicals, total phenolic content, antioxidant activity (DPPH and FRAP), and alpha glucosidase inhibitors. The results of the phytochemical test showed that the extract containing n-hexane contained saponins, steroids/terpenoids and phenols, ethyl acetate and methanol alkaloids, saponins, steroids/terpenoids and phenols. The total phenolic in the n-hexane, ethyl acetate and methanol extracts were 4702.24 ppm, 25862.25 ppm and 21161.03 ppm respectively. The results of the analysis of the antioxidant activity of the extract of Sargassum sp. extracted with n-hexane, ethyl acetate, and methanol using the DPPH method with IC50 values of 524.40 ppm, 329.42 ppm, and 342.15 ppm respectively. The use of the FRAP method showed that the IC50 of each extract (n-hexane, ethyl acetate and methanol) was 318.81, 195.15 and 294.88 ppm. The results of the analysis of alpha glucosidase inhibitory activity inhibition respectively from n-hexane extract (12.64-3.06), ethyl acetate (6.00-2.12 ppm), and methanol (9.40-0.35 ppm). Sargassum sp. extract contains alkaloid, phenolic, saponin and terpenoid compounds. The highest phenolic thiotal content was ethyl acetate extract (25862.25 ppm). The highest antioxidant activity in both methods (DPPH and FRAP) was in ethyl acetate extract with IC50 329, 42 and 195.15 ppm. The inhibition of the alpha glucosidase enzyme of Sargassum sp extract was very weak.
