Cycle threshold value (Ct value) is the number of cycles experienced by the fluorescent signal to reach the threshold in the RT-PCR test which refers to the number of cycles required to amplify viral RNA to reach detectable levels. This study aims to compare the Ct value of patients with confirmed COVID-19 based on vaccination status. The research design is a comparative descriptive with a retrospective approach. Samples were taken using the purposive sampling method during the inspection period from March 2021-September 2022 with a total of 294 samples that met the inclusion and exclusion criteria. The research data consisted of rtRT-PCR results, age, sex, and vaccination history of confirmed COVID-19 patients which were analyzed using descriptive (univariate) and bivariate tests using the Kruskal-Wallis test. The results showed that the most confirmed patients with COVID-19 were in the age group of 18-59 years (92.5%) and the gender with the most confirmed COVID-19 was women (66%). Most of the confirmed COVID-19 patients were those who received 2 doses of vaccination (56.8%). The median Ct values of RdRP and Gene E of patients confirmed with COVID-19 were 19 and 20, respectively. The lowest Ct values for RdRp and Gene E were 10 and the highest was 37. There was no significant difference between the Ct values of RdRP and Gene E on status vaccination with p-values of 0.710 and 0.614 respectively
