Analisis Sekuens Nucleotide Binding Site- Leucine Rich Repeat (Nbs-Lrr) Pada Tanaman Jarak Kepyar (Ricinus COMMUNIS L.)
ABSTRACT
Castor (Ricinus communis L.) is a vegetable oil source plant whose production has decreased every year due to limited resistant cultivar. Grey mold disease caused by pathogenic fungi Botryotinia ricini is the main disease castor. The R gene generally encodes nucleotide binding site-leucine rich repeats (NBS-LRR). The aim of this study was to analyze the NBS-LRR sequence of castor plant in the Agroforestry area of the Meranti Islands KPH (Kesatuan Pengelolaan Hutan). This study used ten samples of castor consisting of five infected plants showing disease symptoms (S1, S2, S3, S4 and S5) and five uninfected plants (R1, R2, R3, R4 and R5). Genomic DNA were isolated from young leaves and PCR was performed using RGA primers and followed by sequencing. The results of this study indicated that the RGA primer can amplify uninfected plant while samples that are infected cannot be amplified. Sample R4 has conserved domain NB-ARC region and PLN03210. In addition, there were 2 motifs namely Kinase-2 and Kinase-3/RNBS-B. Plants that are not infected with the disease have properties that are resistant to grey mold disease they can be used for further studies.
