ON THE MCP-1 EXPRESSION IN THE KIDNEY OF RATS

The kidney is a vital organ that plays an important role in maintaining the body's homeostasis. Provision of a high-fat diet resulted in an increase in oxidized LDL resulting in the proliferation of the glomerular extracellular matrix, then glomerulosclerosis occurred. Sonneratia alba mangrove fruit is a plant that is rich in antioxidants, namely, flavonoids, tannins, saponins, polyphenols, sterols, and alkaloids. This study analyzed the effect of Sonneratia alba fruit extract on MCP-1 expression in the kidneys of rats fed a high-fat diet. The sample used in this study were 16 kidney slide preparations of Rattus norvegicus rats which were divided into 3 groups: the standard group (KS) which was given a normal diet, the positive control group (KP) which was given a high-fat diet, and the treatment group (KT) which was given high-fat diet and Sonneratia alba mangrove fruit extract. The effect of giving Sonneratia alba fruit extract was assessed by immunohistochemical examination. The area of MCP-1 expression is calculated in 5 fields of view and expressed in percent. The expression area of MCP-1 was assessed using the Photoshop Image Analysis program. The results showed that the average percentage of rat kidney MCP-1 expression in group S was 2.407 ± 1.276, group P was 4.412 ± 0.936, and group T was 2.667 ± 1.092. Statistical analysis with One Way Anova showed a significant difference (p=0.018). Further analysis using the Post Hoc test showed significant differences in the S and P groups (p=0.039), as well as the P and T groups (p=0.046). Based on these results, it can be concluded that the administration of Sonneratia alba fruit extract was able to inhibit MCP-1 expression in the kidneys of rats fed a high-fat diet.
