Aktivitas Antioksidan Kitosan Pada Cangkang Kepiting Bakau (Scylla Serrata)
ABSTRACT
Most of the mangrove crab shells are just thrown away or sold without being processed, causing environmental pollution and can damage the stability of the ecosystem. Products that can be produced from mangrove crab shell waste are chitosan which functions as a natural preservative, dye absorbent, and antioxidant. This research aims to determine the antioxidant activity of chitin and chitosan, the effect of different deacetylation heating times, and the best heating time. The stages in this research started with: 1) Preparation of crab shell flour, 2) Extraction of chitin namely demineralization and deproteination processes, 3) Extraction of chitosan namely the process of deacetylation, 4) Antioxidant test of chitin and chitosan using the DPPH (2,2-diphenyl-1-pikriohidrazyl) method. The experimental research method was carried out by direct experiment using a completely randomized design (CRD) with various treatment times of 180 minutes (K1), 210 minutes (K2), and 240 minutes (K3). The results of the research on chitin obtained 32.1% yield, 6.49% water content, 25.12% ash content, 20.2% degree of deacetylation and the IC50 value for antioxidant activity was 352.58 ppm. The results of the chitosan study showed that different heating times had a significant effect on the yield, moisture content and ash content of the mangrove crab shell chitosan. The best results for chitosan were obtained from the K3 treatment with a heating time of 240 minutes with a water content of 4.46%, ash content of 15.88% and deacetylation degree of 70.16%, while the IC50 value for antioxidant activity, namely K1, was 684.53 ppm, K2 is 783.30 ppm and K3 is 997.78 ppm. The IC50 value indicates that the antioxidant activity of the chitin and chitosan of mangrove crab shells (Scylla serrata) is classified as weak or inactive.
