Blastocystis hominis is an intestinal protozoan that can infect humans and animals. It is widely distributed and transmitted via the fecal-oral route. The incidence of Blastocystis hominis is high in developing countries, with poor personal hygiene practices, close contact with pets or contaminated food and drink. Children living in crowded settlements with poor understanding of personal hygiene, inadequate sanitation and facilities that are used simultaneously such as in orphanages are at risk of being infected with Blastocystis hominis. This study is a descriptive study with a cross sectional design with the aim of detecting Blastocystis hominis using a modified jone culture method using sheep serum. Samples were feces of children from 11 orphanages in Pekanbaru. Data were presented in the form of numbers and frequency distribution tables. The characteristics of orphan were elementary student’s, age 6-11 years old, and mosly boys. A total of 95 (children) stool samples were examined from 11 (Pekanbaru) orphanages, 63 children (66.3%) were (found to be) positive for Blastocystis hominis using the culture method, all orphanages used refillable gallon drinking, had cats, and some orphanages had chickens and birds as pets. A high incidence of Blastocystis hominis (66.3%) was detected in the feces of orphanage children in Pekanbaru by culture method.
