Identifikasi Protein Pada Isolat Protein Keong Bakau (Telescopium Telescopium) Dengan pH Berbeda
Telescope snail (Telescopium telescopium) is a gastropod with low economic value and can be utilized as an alternative source of animal protein in the form of protein isolate. The process of isolating telescope snail used protein solubility properties with different alkaline and acidic pH treatments. This study aimed to determine the best pH (alkaline and acidic) in the making of isolates and the effect of pH on molecular weight and type of protein in telescope snail protein isolates. The research method carried out descriptively by laboratory experiment consisted of three different levels with 3 repetitions, namely alkaline pH (9; 10; 11) and acidic pH (4.5; 5.5; 6.5). The stages of this study consisted of preparation and shredding of telescope snail, process of making protein isolates with different alkaline and acidic pH. The results showed the percentage of telescope snail body propotion, namely meat (12.78%), offal (16.67%), and shell (70.56%). The yield value of telescope snail meat flour (25.52 ± 0.53%) with the chemical composition of water content (5.27 ± 0.08%), ash content (4.58 ± 0.08%), and protein content (42.88 ± 0.13%). The value of soluble protein obtained by the best pH in alkaline treatment is pH 11 (5.09 ± 0.02 mg/mL) and acid treatment is pH 4.5 (1.90 ± 0.01 mg/mL) with a yield value (5.06 ± 0.06%). Molecular weight of protein profiles were obtained using the SDS PAGE method with the results of pH 6.5 (4 - 191 kDA), pH 5.5 (4 - 44 kDA), and pH 4.5 (4 - 54 kDA). Protein types identification results from the three treatments obtained in the form of peptides.
