Analisis Sekuens Barkode Dna Rbcl Dan Kandungan Fenol Pada Maman (Cleome Gynandra L.) Asal Riau
Maman plant (Cleome gynandra L.) is commonly consumed as a fermented vegetable by the Rokan Malay community in Riau province. Apart from the potential nutritional and ethnopharmacological benefits of maman plant, maman plant from Riau have not been studied much so information about this species is still difficult to obtain. The purpose of this study was to analyze the rbcL DNA barcode sequence and measure phenol content in maman plant from Riau. The stages of rbcL DNA barcode sequence analysis in maman plant consist of sample preparation, DNA isolation using the CTAB modification method, total DNA electrophoresis, nanodrop spectrophotometry, DNA amplification, amplicon electrophoresis, sequencing and data analysis using BioEdit, BLASTn and Mega version 6 programs. The stages of phenol content analysis consist of sample preparation, extraction, total phenol content test using the Folin-Ciocalteu colorimetric method and data analysis using the SPSS statistical program version 17. The results of molecular analysis showed that the size of the rbcL DNA sequence of maman plants aligned was 833 nucleotides consisting of 738 conserved nucleotides and 95 variable nucleotides. One of the 95 variable nucleotides is the critical nucleotide which ranks 7th. The results of the analysis of phenol content in maman leaf extract from Riau dried by freeze-dried, oven-dried and room air-dried methods were 5.90 ± 0.85 mgGAE/g, 5.14 ± 0.80 mgGAE/g and 4.50 ± 0.73 mgGAE/g. Based on the results of this study, it can be concluded that Gynandropsis gynandra is a species that has the highest percentage value of similarity (99.88%) with maman plants from Riau and the highest total phenol content in maman leaf extract was produced by the freeze-dried method.
