Analisis Sekuens Barkode Dna Its Dan Kandungan Fitokimia Berdasarkan Metode Lc-Ms Pada Maman (Cleome Gynandra L.) Asal Riau
DNA barcoding is a technique used to identify, using one or several standard DNA regions, called DNA barcodes, organisms. Maman is plant that grow and widespread in Rokan Hulu and Rokan Hilir regencies, Riau Province. People in this area often consume this plant in the form of fermented vegetables. Phytochemical characterization is an important source of data for the understanding of plant biodiversity. One method of phytochemical characterization is LC-MS. This study aims to analyze DNA barcode sequences in the Internal Transcribed Spacer (ITS) region and phytochemical content based on the LC-MS method in maman (Cleome gynandra L.). The stages of research were divided into two, namely molecular analysis and phytochemical analysis. The stages of molecular analysis were sampling, total DNA isolation, total DNA electrophoresis, PCR, PCR product electrophoresis, sequencing, and data analysis using BioEdit, BLASTn, and MEGA software version 6.0. The stages of phytochemical analysis were sampling and extraction, and then the sample is analyzed using the LC-MS system. The resulting chromatogram was analyzed using the MassLynx program to obtain the compounds contained in the sample. The determination of compound names was conducted based on the Chemspider database, and the class of compounds was based on PubChem. The length of the maman ITS DNA sequence, was 683 pb. There were 157 nucleotide variations among the accessions studied, and 16 of them were critical nucleotides. In this study, maman samples were very similar to G. gynandra species or its synonym, C. gynandra, with 100.00% identity. The results of phytochemical analysis, namely the total metabolite compounds of maman methanol extract samples detected by LC-MS, were 229 compounds. These compounds belong to the group of alkaloids, phenols, flavonoids, amino acids, carboxylic acid, fatty acids, polyphenol, and steroids. There are eight compounds that have 100% abundance, but only five compounds have been identified by name. Compounds that have 100% abundance were: DL-Phenylalanine (amino acid); N-Boc-1,11-diamino-3,6,9-trioxaundecane oxalate salt (alkaloids); Acetyl tributyl citrate (carboxylic acid); Pheophorbide A (amino acid); and Stigmasterol (steroids).
