Analisis Sekuens Barkode Dna Rbcl Dan Its Pada Tumbuhan Pucuk Seminyak (Champereia Sp.) Asal Riau
ABSTRACT
DNA barcode is a short sequence of a gene positioned within the standardized genome that can identify all species of organisms. Seminyak shoot vegetables are plants originating from Riau, which are one of the traditional Malay vegetables of Rokan. This research aims to analyze the rbcL and ITS gene sequences of seminyak shoot vegetables. The research stages include sampling, isolation using Genomic DNA Mini Kit plant (Geneaid), total DNA electrophoresis, PCR using DreamTaq DNA polymerase (Thermo scientific), PCR product electrophoresis, sequencing, and data analysis using BioEdit version 7.0, BLASTn program, and Mega version 6.0. The research results show that the rbcL sequences from three individuals of seminyak shoots have a size of 836 bp, and the ITS sequences from two individuals of seminyak shoots have a size of 696 bp. Seminyak shoots have yellowish-green flowers and round orange fruits. From the rbcL sequences, there are 26 nucleotide variations and 1 critical nucleotide, while from the ITS sequences, there are 327 nucleotide variations and 3 critical nucleotides. This research on seminyak shoots has a similarity with Champereia manillana with 99.88% identity from the rbcL sequences and has a similarity with Champereia manillana isolate TW3 and isolate TW1 with 95.85% identity from the ITS sequences. In this research, no identity was found to be 100%. The dendrogram results show that this research forms one group with Champereia manillana. These sequences are DNA sequences from the rbcL and ITS genes first reported in seminyak shoots originating from Riau.
