Profil Pita Scar (Sequence Characterized Amplified Region) Terkait Resistensi Terhadap Fusarium Oxysporum F. Sp. Cubense Tropical Race 4 Dan Karakter Morfologi Pisang Di Kecamatan Pangkalan Kerinci Kabupaten Pelalawan
Banana (Musa sp.) is a commodity frequently consumed by people because of its price and easy to obtain. The production of banana has been decreasing mainly due to Fusarium wilt disease caused by pathogenic fungi Fusarium oxysporum f. sp. cubense (Foc) strain tropical race 4 (Foc TR4). This study aims to analyze SCAR (Sequence Characterized Amplified Region) band profile related to Foc TR4 resistance and morphological characteristics on five genotypes of bananas grown in Pangkalan Kerinci District. Morphological observations were evaluated based on the Banana Descriptor and PCR using two SCAR primers (ScaS0901 and ScaU1001) followed by PCR product sequencing. Morphological data were analyzed using the Principal Component Analysis (PCA) method. Morphological data showed that the three major components (PC1, PC2, PC3) produced 87,707% variance. The three morphological characteristics that contribute to the diversity are petiole margins, peduncle length, and fruit shape. PCR results showed that five genotypes of bananas were successfully amplified using both SCAR primers. This indicates that the five genotypes of bananas in this study potentially have resistant traits to Foc TR4. The sequencing results show that the sequences obtained are similar to the amplified sequence using the same two primers deposited in the genebank. Result of this study suggested to provide information about local banana germplasm in the Pangkalan Kerinci District that demonstrate potentially resistant traits against the Foc TR4.
