Analisis Sekuens Barkode Dna Matk Dan Trnl-Trnl-Trnf Intergenic Spacer (Igs) Pada Tumbuhan Pepina (Pterococcus Corniculatus) Asal Riau
Pepina (Pterococcus corniculatus) is one of the native plants of Riau consumed as vegetables by the Malay Rokan community due to its high protein and ascorbic acid content. The pepina plant can be identified using short-length DNA sequences (barcode). The DNA barcodes that can be used are matK and trnL-trnL-trnF Intergenic Spacer (IGS). This study aims to analyze the DNA barcode sequences of matK and trnL-trnL-trnF Intergenic Spacer (IGS) in pepina plants. Pepina samples were collected from Perumahan UNRI Street, Simpang Baru, Tuah Madani District, Pekanbaru City, Riau Province. The research method used was total DNA isolation, Polymerase Chain Reaction (PCR), electrophoresis, sequencing, and data analysis using BioEdit version 7.0, BLASTn, and MEGA 6. The results of the study show that the length of the DNA sequence with the matK DNA barcode in pepina plants is 873 base pairs (bp) and closely related to Plukenetia corniculata species (99.77%). Thirty-three nucleotide variations were found with 2 critical nucleotide variations. Meanwhile, the length of the DNA sequence of pepina plants with the trnL-trnL-trnF Intergenic Spacer DNA barcode is 1067 bp and most closely related to Plukenetia volubilis species (95.22%) and Plukenetia penninervia (94,22%). Two hundred eighty-nine nucleotide variations were found with 13 critical nucleotide variations. In this study, no species with a 100% identity percentage were found. The highest identity percentage was only 99.77% (matK) and 95.22% (trnL-trnL-trnF Intergenic Spacer). This indicates that the pepina DNA sequence is not yet available in the GenBank database.
