Gen 12s Rrna Pada Ikan Baung (Hemibagrus Hoevenii, Bleeker 1846) Di Sungai Kampar, Kecamatan Langgam, Kabupaten Pelalawan, Provinsi Riau
The Kampar River and its tributaries play an important role and have been utilized by communities around the river flow. One of the species of fish that lives in the Kampar River is Hemibagrus hoevenii. Genetic diversity research using the 12S rRNA gene of H. hoevenii baung fish from Kampar River, Langgam District, Pelalawan Regency, Riau Province has never been conducted. The advantages of this gene are that it has an important role in the process of protein synthesis, has a conservative sequence structure and has a high degree of variation. This study aims to analyze the sequence of 12S rRNA in H. hoevenii baung fish originating from Kampar River, Langgam District, Pelalawan Regency, Riau Province. The stages of research carried out are total DNA isolation, total DNA electrophoresis, PCR (Polymerase Chain Reaction), PCR product electrophoresis, sequencing, and data analysis. The results of total DNA isolation were amplified by PCR technique using specific primers for the group genera of Ompok 12SF and 12SR. The annealing temperature used in this study was 58,8oC. The alignment results obtained 416 bp nucleotides. The baung fish H. hoevenii has the highest phylogenetic similarity (87%) with Hemibagrus sp. KR1 and lowest with Ompok hypophthalmus.
