Catfish (Pangasius hypophthalmus) is one of the leading types of consumption fish that is widely cultivated. To meet the needs of catfish, it is necessary to increase cultivation by utilizing empty ponds. Cultivation of catfish in brackish water can cause stress in fish. Feeding with the addition of guava leaf extract to catfish is expected to reduce stress and this can be seen in hematological and physiological catfish.
This study aims to obtain a dose of guava leaves (P. guajava L) for hematological and physiological catfish (P. hypophthalmus). This research was conducted in December 2022 - February 2023. Fish rearing, hematological analysis and fixation of histology organ samples were carried out at the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine Sciences, Riau University. Preparation of gills and intestines of catfish was carried out at the Bogor Agricultural University.
The method used in the study was an experimental method by applying a one-factor Completely Randomized Design (CRD) with five treatment levels, to reduce the level of error, three replications were carried out so that 15 experimental units were needed. The fish used were catfish (P. hypophthalmus) measuring 6-8 cm, reared for 30 days and tested for 14 days. The feed given during maintenance was added with guava leaf extract at a dose of 25, 50, 75, 100 mL/Kg of feed. Sampling was done three times, namely before the fish were treated, day 30 and day 45. Parameters measured were blood profile (number of erythrocytes, leukocytes, hematocrit, hemoglobin and blood glucose), phagocytosis activity, absolute weight growth, feed efficiency, phytochemical test, water quality measurement and histology.
The results showed that in general the addition of guava leaf extract to the feed affects the hematological and physiological of catfish. The condition of fish fed with the addition of guava leaf extract at a dose of 75 ml/kg of feed shows the results of total erythrocytes of 2.40 x 106 cells/mm3, hematocrit value of 35.66%, hemoglobin level of 9.61 g/dL, total leukocytes 10.54 x 104 cells/mm3, blood glucose level of 78.00 mg / dL and phagocytosis activity of 35.66%. Absolute weight growth was 17.76 g, feed efficiency was 41.07%. Fish gill tissue structure was damaged such as hypertrophy, hyperplasia, twisted lamella and broken lamella while in theintestine such as hemorrhage, necrosis and edema.
