Keanekaragaman Pisang Di Kecamatan Tenayan Raya Kota Pekanbaru Berdasarkan Karakter Morfologi Dan Penanda Scar Terpaut Resistensi Fusarium Oxysporum F. Sp. Cubense Tropical Race 4
Bananas are widely utilized since they contain carbohydrates and vitamins. Banana production in Indonesia is threated by plant diseases, especially Fusarium wilt. Fusarium wilt is a soil-borne disease caused by infection from Fusarium oxysporum f.sp. cubense. Identification of resistant bananas to Fusarium wilt disease can be analyzed using molecular markers SCAR (Sequence Characterized Amplified Region). The aim of this study is to analyze the diversity of bananas in Kecamatan Tenayan Raya, Pekanbaru City based on the resistance characteristics to Fusarium oxysporum f. sp. cubense tropical race 4 using the SCAR molecular marker and morphological characteristics. The research method conducted includes morphological observations, total DNA isolation, and Polymerase Chain Reaction (PCR) using two SCAR markers, ScaS0901 and ScaU1001. Principal component analysis (PCA) results of morphological characters shows that lilin banana, barlin banana, and badak banana are located in the same quadrant, indicating high similarity among these three types of bananas. Raja and tanduk are located in a different quadrant from the other bananas. PCR result showed that four banana genotypes (lilin, barlin, badak, and tanduk) were amplified by ScaS0901 and ScaU1001 primers with size of DNA bands about 1,400 bp and 1,600 bp, respectively. However, raja banana was only amplified by marker ScaS0901, indicating that the raja banana has partial resistance.
