Keanekaragaman Pisang Di Kecamatan Marpoyan Damai Kota Pekanbaru Berdasarkan Karakter Morfologi Dan Penanda Scar Terpaut Resistensi Terhadap Fusarium Oxysporum F. Sp. Cubense Tropical Race 4
Banana production tend to decline, particularly due to Fusarium wilt disease caused by Fusarium oxysporum f. sp. cubense tropical race 4 (Foc TR 4). Fusarium wilt is a soil-borne disease causing plant to wilt, discoloration of the pseudostem, and finally death of the whole plant. One of strategies that can be taken is by providing banana germplasms that is resistant to Foc TR 4 disease. This study aims to analyze the diversity of bananas based on Foc TR4 resistance traits used SCAR (Sequence Characterized Amplified Regions) molecular markers and morphological traits. Samples were obtained from Kecamatan Marpoyan Damai, Pekanbaru City, Riau Province. Research methods were morphological observation, DNA isolation, Polymerase Chain Reaction (PCR), and electrophoreses. Principal Component Analysis (PCA) of morphological data was analyzed using Past 4.03. software. Results showed that three principal components with the most contributed characteristics were male bud shape (0.61806), style shape (0.6558), and leaf blade length (0.51263). In addition, four banana genotypes (jantan, raja nangka, serendah, and serawak) produced ScaS0901 and ScaU1001 markers, while mas 40 hari genotype was amplified only by ScaS0901 primer. This result indicate that the four genotypes were potentially resistant to Foc TR4 and the last genotype was partially resistant to Foc TR4. Results of this study can be used as useful data for further studies related to molecular genetic of resistance against Fusarium wilt disease.
