Daya Reduksiagarrumputlautmerah (Gelidium Spinosum) Terhadap Kolesterol Secara In-Vitro
Gelidium spinosum is one type of red seaweed that is beneficial for human health. Gelidium spinosum is one of the raw materials that produce agar. Agar is a complex polysaccharide hydrocolloid composed of repeating disaccharides with units of (1,3)-linked β-D-galactose and (1,4)-linked 3,6 anhydro-α-L-galactose. These polysaccharides cannot be digested in the human intestine directly, so they can function as dietaryfiber. This study aims to determine the cholesterol reduction power in vitro. The method used in this research is an experiment with a non-factorial Completely Randomized Design (CRD), with 3 levels of treatment, namely the addition of agar with different concentrations (0.5 g; 1 g; 1.5 g) and as a positive control, the cholesterol drug Simvastatin. The analysis parameters in this study consisted of Clean Anhydrous Weed (CAW), impurieties, chemical composition of dried G. spinosum, physico-chemical characteristics of agar, antioxidants and reducing power of agar against cholesterol. The results showed that CAW and impurities produced 69.98% and 27.32%. The chemical composition of G. spinosum consisted of 1.25% fat, 8.94% ash, 14.42% water, 17.27% protein and 72.54% carbohydrate, respectively. The physico-chemical characteristics of G. spinosum agar were yield of 25.50%, moisture content of 7.30%, viscosity of 26.8 cP, and sulfate content of 2.83%. Total dietary fiber content was 11.25%, as well as soluble 0.47%, and insoluble 10.78%. Antioxidant activity of G. spinosum agar using the CUPRAC method resulted in an IC50 of 219.8 ppm. Cholesterol reducing power showed that the different amount of agar used can significantly reduce cholesterol. The use of agar as much as 0.5 g; 1 g; 1.5 g can reduce cholesterol by 36.92%, 42.50%, and 47.18% respectively. The results showed that the more the amount of agar to reduce cholesterol, the higher the percentage produced.
