ANALISIS PLASMA DARAH MENCIT JANTAN DEPRESI YANG DIINJEKSI EKSTRAK ETIL ASETAT BIJI PINANG TUA (Areca catechu L.) MENGGUNAKAN LC-HRMS
Depression is a mental illness that can be treated with antidepressant therapy. The extract of natural compounds that have been confirmed to have the potential as antidepressants is areca nut extract which has been tested in vivo through the FST (Forced Swim Test) method of simultaneous double test animals. The test results showed that EEAPT (Ethyl Acetate Extract of Mature Areca Nut Seeds) at a dose of 100 mg/kg was the best dose as an antidepressant and there was no indication of toxic effects during sub-chronic treatment. Further research was conducted with the aim of analyzing the effect of EEAPT antidepressant activity molecularly by looking at the regulation of metabolites that affect depression. There were three treatments carried out in this study, namely depressed mice injected with 100 mg/kg EEAPT, depressed mice injected only with 0.9% saline as a control (-) and healthy mice as a control (+). Mice blood plasma was analyzed using LC-HRMS QTOF, then data processing was carried out with MS-DIAL software to generate the LC-HRMS data results to produce 20.443 molecular features as metabolites. Then a manual filtering process was carried out to remove noise or duplicated molecular features and determine the potential metabolomic profile and 31 putative metabolites were obtained with a high level of confidence identification. Furthermore, a literature review was carried out on the metabolomic profile found and 3 potential metabolites were obtained as peripheral biomarker candidates in depression conditions. Then a multivariate statistical analysis was carried out to see the separation and correlation of metabolites between treatments. The identified potential biomarker candidates came from the lipid metabolism pathway, namely lactosyl ceramide (LacCer), phosphotidylethanolamine (PE) and lysophosphatidylcholine (LPC). However, the metabolite that gave positive results was PE because the ethyl acetate extract of mature areca nut seeds had an immobility reduction time percentage of 17.77%.
