Background: Mesenchymal stem cells (MSCs) have great potential in regenerative medicine applications. Vitamin D has immunomodulatory roles through the interaction with vitamin D receptor (VDR) which is expressed by MSC. The purpose of this study was to determine the effect of vitamin D preconditioning on biological characteristics and expression of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and vascular endothelial growth factor (VEGF) of MSCs.
Methods: Mesenchymal stem cells isolated from umbilical cord tissue and preconditioned with vitamin D3 concentrations of 0-100 nM for 24 and 48 hours were analyzed for viability by CCK-8 assay, while SPM migration analysis with vitamin D3 preconditioning of 50 nM for 24 and 48 hours was assessed by Scratch wound healing assay. In addition, mRNA expression of TNF-α, IL-6, and VEGF after vitamin D3 preconditioning 50 nM for 48 hours was also measured by quantitative real-time Polymerase Chain Reaction method.
