Hidrolisis Protein Teripang Emas (Stichopus hermanii) Menggunakan Enzim Papain Dengan Konsentrasi Berbeda
Hydrolysis is a process of degrading proteins with acids, bases, or proteolytic enzymes to obtain amino acids and peptides. Golden sea cucumber (Stichopus hermanii) is a fishery commodity with good prospects and high economic value in domestic and international markets. The hydrolysis of golden sea cucumber involved the use of papain enzyme as a catalyst for protein breakdown. The advantages of using papain in this research included its easy availability, lack of side reactions, non-toxicity, relative heat stability, and high catalytic activity. Due to the many uses of papain enzyme in this research, therewas an increasein demand for papain in the food industry. The purpose of this study was to determine the proportion analysis of golden sea cucumber, the chemical composition of golden sea cucumber flour, the effect of adding papain enzyme at different concentrations, analyzing the best degree of hydrolysis value, and analyzing dissolved protein in the protein hydrolysate of golden sea cucumber flour with the addition of papain enzyme at different concentrations.The method used in this research was an experimental method, which involved performing hydrolysis of golden sea cucumber protein using papain enzyme at different concentrations. The design employed was a completely randomized design with a non-factorial approach. The treatments consisted of three levels of hydrolysis using papain enzyme at concentrations of 4%(P1), 5%(P2),and 6%(P3). Theresults showed that thehighest proportion of fresh golden sea cucumber was the meat, accounting for 53.33%, followed by the skin at 13.33%, and the viscera at 33.34%. The chemical composition of the golden sea cucumber meat flour included a moisture content of 6.46%, an ash content of 18.73%, and a protein content of 56,23%. The best hydrolysis result for golden sea cucumber protein hydrolysate was achieved at a concentration of 6%, with a hydrolysis degree calculation of 17.04%. The best soluble protein test result for the sea cucumber hydrolysate was obtained at a concentration of 6%, with a value of 2.10 mg/mL.
