Isolasi Dan Karakterisasi Bakteri Pelarut Fosfat Dan Kalium Dari Tanah Rhizosfer Tanaman Padi
Rhizosphere bacteria are bacteria found in plant roots and have high diversity. Rhizosphere bacteria for plants play a role in increasing the availability of phosphorus and potassium in the soil through the mechanism of dissolving organic phosphate and potassium into soluble forms This study aims to isolate, characterize and test the phosphate and potassium dissolution of rhizosphere bacterial isolates of rice plants in Pembatang Village, Pangean District, Kuantan Singingi Regency, Riau. Characterization of phosphate and potassium solubilizing bacteria isolates included macroscopic, microscopic and catalase tests. The results obtained five isolates of phosphate solubilizing bacteria namely BPF 1.1, BPF 1.4, BPF 2.1, BPF 3.1 and BPF 3.2 which were isolated using Pikovskaya medium with phosphate source Ca3(PO4)2 and seven isolates of potassium solubilizing bacteria namely BPK 1.1, BPK 1.2, BPK 2.1, BPK 2.2, BPK 2.3, BPK 3.1 and BPK 3.2 using Alexandrov medium with potassium feldspar source (KAISi3O8). The results of Gram staining obtained phosphate solubilizing bacterial isolates are Gram positive one isolate is BPF 3.2 and Gram negative four isolates are BPF 1.1, BPF 1.4, BPF 2.1, and BPF 3.1. The results of Gram staining of potassium solubilizing bacteria obtained Gram positive bacterial isolates of one isolate namely BPK 3.2, Gram negative six isolates namely BPK 1.1, BPK 1.2, BPK 2.1, BPK 2.2, BPK 2.3 and BPK 3.1. Catalase test results were positive for four phosphate-solubilizing bacterial isolates and seven potassium solubilizing bacterial isolates. The results of phosphate solubilizing activity of isolate BPF 1.4 with the highest phosphate solubilizing index of 2.33 and potassium solubilizing activity of BPK 3.2 with the highest potassium solubilizing index of 2.48 with Alexandrov media source of potassium feldspar (KAISi3O8) and isolate BPK 2.1 with the highest potassium solubilizing index of 2.34 with potassium source K2HPO4.
