Gen 16s Rrna Ikan Hidung Budak (Ceratoglanis Scleronema) Dari Sungai Kampar Kecamatan Langgam, Kabupaten Pelalawan, Provinsi Riau
Kampar River is a river that also has various types of fish that are popular with the community and are fish that are characteristic of flood-prone areas. One type of fish is the hidung budak fish (Ceratoglanis scleronema). Research on genetic diversity using the 16S rRNA gene of hidung budak fish C. scleronema from the Kampar River, Langgam District, Pelalawan Regency, Riau Province has never been done. This study aims to analyze the 16S rRNA sequence in hidung budak fish C. scleronema from the Kampar River, Langgam District, Pelalawan Regency, Riau Province. The stages of the research carried out were total DNA electrophoresis, Amplification with PCR (Polymerase Chain Reaction) Technique, PCR product electrophoresis, sequencing, and data analysis. The results of total DNA isolation were amplified using PCR (Polymerase Chain Reaction) technique using universal primers for the genus Ceratoglanis 16SF (Forward) and 16SR (Reverse). The annealing temperature used in this study was 58.8oC. The results of the 16S rRNA sequence alignment in the hidung budak fish C. Scleronema (Cs1K, Cs4K and Cs5K) obtained 452 bp. This proportion covers 45.2% of the expected 1000 bp. The results of the BLASTn analysis showed that C. scleronema (Cs1K, Cs4K, and Cs5K) and Ompok eugeneitatus have an identity percentage of 94.43% and Ompok hypophthalmus 99,44%.
