Analisis Sekuens Nbs-Lrr Ramin (Gonystylus Bancanus (Miq.) Kurz) Yang Tumbuh Di Hutan Rawa Gambut Provinsi Riau
Gonystylus bancanus (Miq.) Kurz (ramin) is an endemic tree species of tropical peat swamp forests and is currently classified as critically endangered species. Disease resistant genes such as genes encoding NBS-LRR protein are important to maintain the existence of the species. This study aimed to analyze the NBS-LRR gene sequence in ramin growing in the production forest of PT Diamond Raya Timber, Riau Province. Three samples of young ramin leaves were taken for extracting total genomic DNA. PCR results using RGA primers showed DNA band of approximately 250 bp in all three ramin samples. The deduced amino acid sequences revealed NB-ARC conserved domain. NBS motif (P-loop) was found in all samples. This motif indicates ATP-binding functionality essential for plant immune activation. BLASTp analysis indicated that the deduced amino acid sequences shared similarity with disease resistance RPP13-like protein 4 from Pistacia vera, Citrus sinensis, and Rosa chinensis, showing 41–50% identity and E-values
