Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Ekstrak Etil Asetat Ranting Taxus Sumatrana
Taxus sumatrana is a plant that grows in the highlands and is included in the Gymnospermae subdivision. The branches of the T. sumatrana have various chemical contents with several biological activities. According to various studies, T. sumatrana contains secondary metabolites such as alkaloids, saponins, flavonoids, and terpenoids. Secondary metabolite extracted from T. sumatrana are know to possess biological activities such as anti-cancer, anti-diabetes, anti- tumor and antioxidant. The objective of this research is to determine secondary metabolites of ethyl acetate extract of T. sumatrana twigs and conduct toxicity tests using the BSLT (brine shrimp lethality test) method on pure compounds of T. sumatrana twigs. The research employed the maceration method with ethyl acetate as a solvent to obtain the plant extract. The extract was then separated using vacuum liquid chromatography (VLC), resulting in 12 fractions. The fractions obtained were subjected to thin layer chromatography (TLC) and a dark brown compound weighing 12 mg. The purity of this compound was analyzed using thin layer chromatography (TLC) and high- performance liquid chromatography (HPLC). The compound was designated TS-SFG3-F and characterized using a UV-Vis and FTIR spectrophotometer. The UV-Vis spectrum showed maximum absorption at wavelengths of 229.5 nm and 278.5 nm, with absorbance values of 2.38 and 1.06, respectively. The FTIR spectrum shows that the TS-SFG3-F compound has several absorption bands at the wave numbers of 3364 cm-1 (O-H), 2924; 2852 cm-1 (aliphatic C-H), 1718 cm-1 (C=O), 1612 cm-1 (C=C), 1449 cm-1 (C=N), 1373 cm-1 (aliphatic CH), 1149;1035 cm-1 (C-O), dan 822;716 cm-1 (aromatic CH). Based on this study, it can be inferred that TS-SFG3-F is a flavonoid. The results of the toxicity test using the BSLT method showed that compound TS-SFG3-F was toxic with a LC50 value of smaller 10 ppm.
