Pengaruh Pemberian Naa Dan Kinetin Terhadap Pertumbuhan Eksplan Buah Naga (Hylocereus Costaricensis) Melalui Teknik Kultur Jaringan Secara In Vitro Sebagai Sumber Belajar Pada Konsep Bioteknologi Bagi Siswa Sma
This research aimed to determine the effect of Naphtalene Acetic Acid (NAA) and Kinetin on the growth of explants dragon fruit (Hylocereus costaricensis) as a learning resource on the concept of biotechnology for high school students. This research was conducted at the Laboratory of Biology Education University of Riau and Laboratory Biotechnology of Riau Islamic University from May to August 2012, using a completely randomized design of 3 x 3 factorial with four replications. factor first (N) is NAA consists of 3 level: 0 ppm, 0.2 ppm, 0.4 ppm. K factor of two is Kinetin which consists of 3 level: 0 ppm, 3 ppm, 4 ppm. parameters observed were percentage grows, the number of shoots, shoot height and number of roots were analyzed by ANAVA and tested further by DMRT at 5% level. Whereas for the current parameters appearing callus, as it appears shoots and roots emerge when analyzed descriptively. These results demonstrate that administration of NAA and Kinetin significantly affect the growth of explants Dragon Fruit. Percentage grew 100% explant all treatments except treatment N0K4, N0,2K0, N0,2K3. In this research does not appear callus. Treatment N0K4 able to create the fastest buds at 13 days after planting. Highest amount of buds in the treatment of N0,4K3 is 2.25 buds. While high best buds in treatment N0,4 K0 is 2.45 cm. Treatment N0,2K0 able to bring the fastest root is 10 days after planting. Highest amount of roots in treatment N0,4K4 is 5.25 roots. According to the research treatment N0,4 K4 is the best treatment for the growth of explants Dragon Fruit. Facts–facts the results of this research can be used as a learning resource development in biotechnology concepts for high school students in the form of student worksheets.
