Sensitivitas Kulit Buah Manggis (Garcinia Mangostana L.) Terhadap Bakteri Aeromonas Hydrophila
The mangosteen pericarp juice has been used for medicine as it has antimicrobial agents. However, information of the use of mangosteen juice in fish is almost none. In culture fish, the attack of Aeromonas hydrophila causing the MAS disease that cause big lost. To understand the sensitivity of mangosteen pericarp juice on Aeromonas hydrophila growth, a study has been conducted from July to October 2014. Mangosteen juice was prepared by juicing 9.5 gram of mangosteen pericarp and 50 ml aquadest. The juice was then filtered using a cotton cloth. There were randomized design 11 treatments 3 replications different mangosteen dosages applied for sensitivity test, namely P1 (19 g/L), P2 (38 g/L), P3 (57 g/L), P4 (76 g/L), P5 (95 g/L), P6 (114 g/L), P7 (133 g/L),P8 (152 g/L), P9 (171 g/L) dan P10 (190 g/L) and the parameter measured was the clear zone size. The sensitivity of mangosteen juice in inhibiting the A. hydrophila colony growth that was cultured in TSA was tested using 3 different dosages, there are 57 g/L, 76 g/L and 95 g/L. To understand the LD50 Clarias gariepinus that was treated with the mangosteen juice, 4 treatments applied, namely 1.9 g/L, 3.8 g/L, 5.7 g/L and 7.6 g/L. Results indicate that start form the dose of 95 g/L the mangosteen juice was effective to inhibit the growth of A. hydrophila (13.65 mm clear zone and 30.6 X 108 cfu/mL colonies). The LD50 of the C. gariepinus, however, was achieved in the dose of 4.43 g/L, far below the sensitive dose of the A. hydrophila . Based on data obtained, it can be concluded that the mangosteen juice can not be used for curing the MAS disease in fish, as the lethal dose for fish is lower than the antimicrobial effective dose.
