Isolasi Dan Uji Aktivitas Antidiabetes Senyawa Metabolit Sekunder Dari Ekstrak Metanol Akar Tumbuhan Tunjuk Langit (Helminthostachys Zeylanica)

Helminthostacshys zeylanica is a plant of the family of Ophioglossaceae known as tunjuk langit. The roots of this plant are used as a 100 days cough remedy, dysentery, nasal disease and lung disease. This plant contains saponins and flavonoids. This study aims to isolate and characterize the flavonoid compounds from the roots of this. Plant Isolation methods used was based on polarity fractionation method. Some 7.5 kg of the roots was macerated with 17 L methanol for 3x24 hours, followed by ultrasonication for 1 hour. The some 100 g of dried extract was dissolved in methanol then partifioned with n-hexane (2:1). Methanol fraction was dried then was subjected to chromatography liquid vacuum VLC) to give 12 fractions (n-hexane – ethylacetate – methanol). Flavonoid compound was obtained from fractions 5 and 6. These were combined and subjected to flash chromatography. Flavonoid compound was afforded as yellow crystal in vial 19 and it was coded as HZ19. The purity of this compound was analyzed by TLC and HPLC as well as determination of the melting point. Structure of pure compound was determined using spectroscopy methods including UV-Vis, infrared (IR) and NMR. Based on spectroscopic data HZ19 similar to ugonin group. Antidiabetic test by inhibition of α-glucosidase enzyme (in vitro) found that IC50 value of the compound was 404.992 µg/mL. A compound activity ugonin is lower than acarbose activity (standard drug) which is 1 µg/ mL.
