ABSTRACT
Purple sweet potatoes (Ipomoea batatas) tubers contain phenolic compounds which act as natural antioxidants. Extraction of phenolic compounds from purple sweet potatoes can be done by either one of two methods. The first method is the Pothitirat modified extraction method. In this method,the dry powder of I. batatas was incubated with 50% (v/v) ethanol in Na-acetate buffer pH 5.5 0.05 M at 40°C for 8 days. The second method is the Bridgers modified extraction method. In this method, the dry powder of I. batatas tubers was incubated with 40% (v/v) ethanol, Na-acetate buffer pH 5.5 0.05 M at 40°C for 60 hours, followed by incubation with 70% acidic methanol (pH 3.5) for 1 hour. All extractions were done with and without the assistance of Trichoderma asperellum LBKURCC1 cellulase. The activity of the T.asperellum LBKURCC1 crude cellulase used for the enzyme assisted extraction was (4.1 ± 0.7) U/mL. The total phenolic concentration obtained in the I. batatas tuber extracts were determined by the Folin-Ciocalteau method. The results showed that the Pothitirat modified extraction method gave significantly lower total phenolic concentration in extracts than the Bridgers modified extraction method (p˂0,05). The total phenolic concentration in water extracts without cellulase assistance by the Pothitirat modified extraction method was (24.7 ± 0.7) mg/g of dry extract, while that by the Bridgers modified extraction method was (28.7 ± 0.8) mg/g of dry extract. The total phenolic concentration in water extracts with cellulase assistance by the Pothitirat modified extraction method was (26.9 ± 0.8) mg/g of dry extract, while that by the Bridgers modified extraction method was (17.8 ± 0.4) mg/g of dry extract. The total phenolic concentration in ethanol extracts without cellulase assistance by the Pothitirat modified extraction method was (28.7 ± 0.2) mg/g of dry extract, while that by the Bridgers modified extraction method was (35.7 ± 1.2) mg/g of dry extract. The total phenolic concentration in ethanol extracts with cellulase assistance by the Pothitirat modified extraction method was (25.5 ± 0.5) mg/g of dry extract, while that by the Bridgers modified extraction method was (30.0 ± 0.5) mg/g of dry extract.
