Isolasi Gen Housekeeping Ef1α (Elongation Factor 1α) Pada Tumbuhan Durik-Durik (Syzygium Sp.) Dari Danau Kajuik, Kabupaten Pelalawan, Provinsi Riau
Durik-durik (Syzygium sp.) is one of the important plants grown in floodplain area around Kajuik Lake Langgam District, Pelalawan Regency, Riau Province and has a good adaptability to the inundation stress. This ability is related to the source of the tolerant genes to its inundation stress. It is necessary to do the gene expression study for identifying those genes. In gene expression analysis, internal controls are generally required from housekeeping gene group such as the EF1α gene. This study aims to isolate EF1α gene in Durik-durik plants (Syzygium sp.) from Kajuik Lake. The research procedures included sampling, total DNA isolation, PCR, electrophoresis, sequencing and data analysis. Data analysis was conducted using Bioedit program version 7.0, BLASTn, and MEGA version 7.0. The DNA sequence obtained in this study was true of the gene encoding Elongation factor 1α with sized 837 bp and had similarity of 89-91% with EF1α sequences from other plant species. EF1α primer pair specific for Syzygium sp. has been successfully designed, namely forward 5'- ACA AAT GGA ATT TTG TGG GG-3 'and reverse 5'-GCT GTT CGA TGA CTC TAA CC-3'.
