Isolasi Gen Housekeeping Beta-Tubulin Pada Tumbuhan Durik-Durik (Syzygium Sp.) Dari Danau Kajuik Kabupaten Pelalawan Provinsi Riau
Durik-durik (Syzygium sp.) is one of the living plants and has adapted to the floodplain area around Kajuik Lake, Riau Province. This plant has an important role as a source of nutrients, animal habitats and prevent erosion in the floodplain lake. This study aims to isolate the Beta-tubulin housekeeping genes in Durik-durik plants (Syzygium sp.) from Kajuik Lake, Pelalawan District. The total DNA molecule was extracted using the Genomic DNA Mini Kit Plant (Geneaid), then amplified using universal Beta-tubulin primer. The PCR product was migrated on 1% agarose gel in 1X TBE buffer. BioEdit Programs Version 7.0, BLASTn, and MEGA Version 6.0 are used for data analysis. The results indicated that the DNA sequence of the Beta-tubulin gene had a size of 1167 bp and encoded 311 amino acids of deduction. The Durik-durik gene sequence had 92% -95% similarity with the Beta-tubulin gene sequence in some plant species.
