Identifikasi Molekuler Bakteri Endofit Enterobacter Cloacae Lbkurcc146 dan LBKURCC147
ABSTRACT
LBKURCC146 and LBKURCC147 are cellulolytic endophytic bacteria that can degrade cellulose into simpler compounds such as glucose. These isolates that isolated from Harupat ferns stem in Pekanbaru, Riau are resistant to acidic pH. It was identified morphologically as Enterobacter cloacae bacteria called LBKURCC146 and LBKURCC147 and stored in the Research Laboratory of Enzyme, Fermentation and Biomolecular, at Chemistry Department , Mathematics and Natural Sciences Faculty, University of Riau. Morphological identification diversity level has limitations for valid identify of microbial species, therefore it is necessary to do molecular identification and phylogenetic relationships. DNA of LBKURCC146 and 147 bacteria were isolated after culturing for 24 hours inccubation using Geneaid Kit, then amplified using universal 16S rDNA primers and cloacae cheV primers which have been designed previously. The size of bacterial DNA obtained from the isolation results is above 10037 pb. The size of bacterial DNA after amplification with universal 16S rDNA primers and cloacae cheV primers are around 1400 pb and 500 pb respectively. The analysis with 16S rDNA sequence showed that species of LBKURCC146 and 147 had 99% similarity with several Enterobacter and Bacillus species. The results of the analysis with cloacae cheV sequence confirmed that LBKURCC146 and 147 were E. cloacae with 100% similarity. Based on the result, it is concluded that molecular identification showed in line result with morphological identification.
