Isolasi Dan Seleksi Bakteri Penghasil Biosurfaktan Dari Air Terkontaminasi Ceceran Minyak Di Sumur Pengeboran Minyak Bumi Pt. Bumi Siak Pusako – Pertamina Hulu Desa Kasikan Kabupaten Kampar
Oil spill contaminated water present around the oil well drilling well. The presence of crude oil may trigger gene mutation in specific bacteria present in that area . The bacteria may produce biosurfactant for degrading the oil. A study aims to obtain biosurfactant producing bacteria from oil spill contaminated water around petroleum drilling wells in the PT. Bumi Siak Pusako – Pertamina Hulu Kasikan Village Kampar Regency has been conducted from July to August 2018. There were 2 stations, namely St1 (around the Ksk033 and the Ksk020 wells) and St2 (around the Ksk006 well and the Gathering Station). The water sample was taken to laboratory and 5 ml of the sample was mixed with Enrichment media to grow the bacteria. The bacteria were isolated and purified using SMSSe and Nutrient Agar media. The purified bacteria were then morphologically studied, results shown that there were 6 bacteria present. The bacteria obtained were then analyzed using Oil Spreading Assay (OSA) and Emulsification Index (EI) test. Bacteria isolates present, namely Is1 (circular form, entire margin, raised, OSA +, EI 58%); Is2 (irregular form, umbonate, undulate, OSA +, EI 29%), Is3 (circular form, entire margin, umbonate, OSA +, EI 40%), Is4 (circular, entire margin, raised, OSA +, EI 12%), Is5 (circular, entire margin, convex, OSA +, EI 42,85%), Is6 (rhizoid, filiform, flat, OSA +, EI 18%). Results of these tests indicate that those bacteria were potential for producing biosurfactant. Keywords : OSA, Emulsification Index, Biodegradation, Oil Pollutant
