Analisis Keanekaragaman Genetik Kulim (Scorodocarpus Borneensis Becc.) Di Provinsi Riau Berdasarkan Penanda Molekuler RAPD
This study was aimed to determine the genetic diversity of populations of Kulim commonly grown in Riau Province by RAPD markers. The analysis was carried out at Laboratory of Molecular Genetics, Center of Research and Development of Biotechnology and Tree Improvement of Forest Plants in Purwobinangun, Sleman, Yogyakarta. Cambium from 36 trees were sampled from 3 populations, i.e. from Bengkalis, Kampar and Indragiri Hulu for DNA extraction by using modification of cetyl trimethyl ammonium bromide (CTAB) method. The DNA was then ampified using six RAPD primers (Operon Technology) i.e. OPC-6, OPY-6, OPY-13, OPY-14, OPO-6, and OPW-4. Bands profiles of RAPD were transformed into binary data by scoring existed band with 1 and non-existed band with 0. The data was then further analyzed with POPGENE 1.32 software. The measured variables of genetic diversity within population were percentage of polymorphic loci (PLP), number of effective alleles (Ae), Shanon index (S) and genetic diversity of Nei-s (He). Meanwhile, the variables for genetic diversity among populations were genetic distance (d), genetic differentiation (GST), and group analysis.
A total of 51 loci detected, 47 loci (92.16%) were polymorphic loci. The polymorphic loci in three populations were ranged from 71.79% to 79.49% with a mean of 76.50%±0.04. The average number of effective alleles was 1,359±0.08. Genetic diversity within population (He) was considered as high with a value of 0.226±0.04 and Shanon index of 0.351±0.06. Among the three populations, Kampar exhibited significantly higher variability than other populations. Genetic differentiation among population was fairly low (0.1191). The genetic distance Nei among population ranged from 0.0112 to 0.0825 with mean of 0.0481. The dissimilarity coefficient for all samples was ranged from 0.00 to 0.86, while the grouping analysis showed that sampels grouped into two main clusters with a dissimilarity coefficient of 0.86. The lowest coefficient value (0.00) is dominated by samples of Duri and Indragiri Hulu populations.
