Optimasi Waktu Penambahan Bakteri Elisitor Staphylococcus Aureus Pada Penicillium Sp. Lbkurcc34 Untuk Produksi Metabolit Sekunder Berpotensi Antibiotik

Penicillium sp. LBKURCC34 is a fungus isolated from peat soil in primary forest, Giam Siak Kecil Bukit Batu Biosphere Reserve (GSKBB), Riau Province. This fungus has potency to produce secondary metabolites that have antimicrobial activity. The study aimed to determine the time for addition of biotic elicitor (Staphylococcus aureus) based on analysis of the secondary metabolite content and activity on inhibition of several pathogenic microbes. Determination of the growth curve was carried out by the gravimetric method, which was analyzed from day 0 to day 10 of incubation time. The production of secondary metabolites was carried out by the addition of biotic elicitor (Staphylococcus aureus) on the 2nd day of fermentation (14 days incubation) and the 4th day of fermentation (6 days and 14 days of incubation) that these time was based on the growth curve obtained. After the fermentation incubation the production media and mycelium was separated by vacuum filtration, then extracted using ethyl acetate. The extract was concentrated and reconstituted with methanol. Antimicrobial tests were carried out by disc diffusion method against Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Bacillus subtilis, Candida albicans and dilution methods (MIC and MBC) using Escherichia coli. There were slightly different trends of growth curve of Penicillium sp. And Penicillium sp. with biotic elicitor (Staphyloccocus aureus). KCKT kromatograms on 210 nm and 254 nm showed different number of peak on different retention time. Antimicrobial activity of disc diffusion method revealed the condition were on H4P14 extract against Bacillus subtilis bacteria with a percentage of 56,73% compared to positive controls. The three extracts obtained were not able to inhibit the growth of Escherichia coli bacteria by dilution method. The best time to add elicitor (Staphyloccocus aureus) as stimulation on production secondary metabolite that have antimicrobial activity are adding elicitor on the 4th days and incubation for 14 days.
