Histopatologi Insang, Ginjal Dan Hati Ikan Jambal Siam (Pangasius Hypophthalmus) Yang Diberi Pakan Mengandung Tepung Kunyit (Curcuma Domestica Val.)
This study was conducted from February to Mei 2019 and that locate was Laboratorium Parasit dan Penyakit Ikan. This study aims to determinate the effect of giving turmeric flour on histological structure of Pangasius hypophthalmus. The method in this research was the experimental method using the Completely Randomized Design (CRD) with one factor and five treatment levels and three time replication. Kn: (were not feed with turmeric flour (Curcuma domestica V) and without were infected with Aeromonas hydrophila, Kp: (were not fed with turmeric flour (Curcuma domestica V) and were challenged with Aeromonas hydrophila), P1: fed with turmeric flour (Curcuma domestica V) at dose 0,5 g/kg feed, P2: dose 0,7 g/kg feed, P3: dose 0,9 g/kg feed. Fish reared size 8 – 12 cm and reared in an aquarium the size of 30x30x40 cm with stocking density 1 fish/3 L, feeding do 3 times a day 08.00, 12.00 and 18.00 WIB as many as 10%. Sampling for histological structure was carried out 3 times at the beginning of maintenance, 30 days of maintenance and 14 days after being challenged. The organs observed were gills, kidneys and liver. The organs were fixed with 10% formalin for 24-48 hours and were processed to histology. Samples were cut with a microtome with a thickness of 5 μm. Then, fish organs were histologically processed and stained (HE). Results shown that the structure of gill, kidney and liver of catfish of giving turmeric flour were challenged with Aeromonas hydrophila was normal. The histological structure of gills, kidney and liver of giving turmeric flour slight damage such as hyperplasia, deformasi lamella and lifting epithelium. HAI score 1.33 – 4.33 was category normal. The structure of gills without of giving turmeric flour (Kp) the showed slight damage with score HAI 12.67. The histological structure of kidney of P1, P2, and P3 the showed hypertrophy, hemorrhage and necrosis. The histological of liver of P1, P2 and P3 the showed hypertrophy, degeneration vacuola and necrosis. P2 (feed with turmeric flour (Curcuma domestica V) dose 0,7g/kg feed), were the best treatment. Survival rate of fish was 100%. The water quality was, temperature was 28-29 0C, pH was 6,5-7,0, dissolved oxygen was 4,0-4,3 mg/L, ammonia 0,16-0,50 mg/L.
