Isolasi Dan Uji Aktivitas Antibakteri Ekstrak Akar Tumbuhan Spathoglottis Aurea Lindl
Spathoglottis aurea Lindl plants or orchids are a tribe of Orchidaceae that is
widely available in Indonesia. This plant is commonly known to the public for a long
time, even the roots plants Spathoglottis aurea Lindl used to treat sore eyes for the
people Rokan Hulu. Spathoglottis aurea Lindl root dry powder obtained from Rokan
Hulu, Riau Province macerated with methanol. The methanol extract was partitioned
with hexane several times and the solvent was evaporated with a rotary evaporator to
obtain a total extract methanol and hexane extracts total. The methanol extract and
hexane extract further fractionated using column chromatography gravity. In the
methanol extract made elution with different eluent from 100% ethyl acetate, ratio of
ethyl acetate: methanol up to 100% methanol and the obtained results are 4 fractions.
While elution hexane to extract hexane starting from 100% ratio of hexane: ethyl
acetate up to 100% ethyl acetate and separation 4 fractions obtained results. In the
fraction 2 (Fh2) hexane extract obtained white crystals. Fraction 2 (Fh2) further test
the purity by TLC observation, determination of melting point and characterized by
UV, FTIR and GC-MS. The results GC-MS analysis of unknown compound shaving
the formula C29H50O Fh2 with a peak molecular ion m/z 414 in accordance with the
structure of β-sitosterol. The test results of antibacterial activity that is carried out,
and methanol extract of the roots of plants Spathoglottis aurea Lindl hexane not
active against S. aureus and E. coli bacteria. While the fraction Fh1 and Fh2 hexane
extract active against S. aureus and E. coli bacteria with inhibitory region (DDH) is
about 10-12 mm.
