Uji Ekstrak Etanol Dan Infusa Daun Asam Jawa, Herbal Benalu, Herbal Putri Malu Sebagai Inhibitor Enzim Α-Amilase Dan Α-Glukosidase
Indonesia is one of the country which rich with the plants, around 940 species of that
are medicinal plants. One of them i.e., asam jawa (Tamarindus indica L), benalu api
herb (Scurrula sp) and putri malu herb (Mimosa pudica D) are medicines plants that
always consumed by the society and can be use as the antidiabetic formula, by
inhibition α-amylase and α-glucosidase enzymes. α-amylase and α-glucosidase
enzymes were the enzymes to hydrolyze carbohidrate (starch) become to glucose, and
the glucose will be absorb by the blood. If the glucose over normal (>140 mg/dL) this
is showed that the people diagnoted diabetes mellitus. The objective of this study
were to determine percent inhibition of ethanolis extract and aqueous extract (infusa)
from asam jawa leaves, benalu api herb and putri malu herb in fresh and dired
condition to α-amylase and α-glucosidase enzymes. Acarbose was used as positive
control. Inhibition of α-amylase was determine by DNS methode, and for α-
glucosidase using a p-NPG substrate, with measured the absorbance using
spectrophotometer (microplate reader) at 530 and 410 nm. The result showed that the
plants extract more potential for inhibitied α-glucosidase with average up to 50% and
less potential for inhibitied α-amylase. Percent inhibition of α-amylase that were not
significantly different from acarbose (93.89±0.02) i.e., infusa of benalu api dried herb
(85.58±2.93), infusa of putri malu fresh herb ( 87.40±1.81) and infusa of putri malu
dried herb ( 98.85±0.66), while percent inhibition of α-glucosidase that were not
significantly different from acarbose (97.99±0.19) i.e., infusa of asam jawa dired
leaves (99.10±0.36), infusa of benalu api fresh herb (97.96±0.85) and infusa of putri
malu fresh herb (98.64±1.25).
