Uji Ekstrak Etanol Dan Infusa Bawang Putih, Daun Mangkokan, Temu Giring Sebagai Inhibitor Enzim Α-Amilase Dan Α-Glukosidase
The inhibition process of the digestive enzymes, α-amylase and α-glucosidase can
have the effect to reducing blood glucose levels, especially in type 2 diabetic
patients. So, the objective of the study were to determine in-vitro α-amylase and
α-glucosidase inhibitory activities of 96% ethanolic extracts and infusa (aqueous
extracts) from medicinal plants, i.e., garlic (Allium sativum L.), mangkokan leaves
(Nothopanax scutellarium Merr.) and temu giring (Curcuma heyneana Val. & V)
in their fresh and dried samples. Inhibition of the samples to α-amylase was
determined by DNS method, while α-glucosidase using a p-NPG substrate. The
absorbance was measured using microplate reader at 530 nm for α-amylase and
410 nm for α-glucosidase. Acarbose was used as positive control. The results
showed that all samples are potential inhibitors for α-amylase, which was proven
by 4 extracts that were not significantly different from acarbose (93.89 + 0.02),
i.e., infusa of mangkokan fresh leaves (88.62 + 5.04); infusa of mangkokan dried
leaves (91.64 + 1.17); infusa of fresh temu giring (76.05 + 11.84) and infusa of
dried temu giring (90.46 + 2.67). For α-glucosidase, the samples were
significantly lower than acarbose, with inhibition of acarbose was 97.99 + 0.19.
