Isolasi Gen Housekeeping Gapdh (Gliseraldehid-3- Fosfat Dehidrogenase) Pada Durik-Durik (Syzygium Sp.) Asal Danau Kajuik Kabupaten Pelalawan Provinsi Riau
Durik-durik (Syzygium sp.) is one of the plants that have been adapted to wetland
ecosystem in Kajuik Lake, Langgam District, Pelalawan Regency, Riau Province.
The ability of this plant adaptation can’t be separated from the existence of genes
related to the stagnant puddle adaptation. To know the genes involved in the
stress, a gene expression analysis can be performed. As a first step, it has been
done GAPDH housekeeping gene isolation which can then be validated as a
reference gene. The research procedures included sampling, total DNA extraction,
electrophoresis, PCR, sequencing and data analysis. Data analysis using BLASTn
program, Bioedit Version 7.0. and MEGA Version 7.0. In this study, GAPDH
gene fragment was measured about 1100pb and after sequencing, obtained a
sequence of nucleotides along 1136pb. GAPDH gene sequences from Durik-durik
are 84 - 95% similar with GAPDH gene sequences from some plant accessions.
