Analisis Keanekaragaman Genetik Kacang Hijau (Vigna Radiata L.) Asal Provinsi Riau Berdasarkan Penanda G-Ssr (Genomic-Simple Sequence Repeat)
Mungbean (Vigna radiata L.) is an important legume after soybean and peanut.
This study aime to analyze the genetic diversity of 10 mungbean genotypes from
Riau Province using g-SSR (Genomic-Simple Sequence Repeat) marker. The
DNA total was extracted from fresh leaves then amplified using 8 primer pairs of
g-SSR. The PCR products were migrated on 2,5% agarose gel in 1X TBE buffer.
The DNA bands were converted into binary data such as “0” if the band was
absent and “1” if the band was present. The binary data was then computed using
NTSYS-pc version 2.02i by UPGMA method. The results showed that the
similarity coefficient of 10 genotypes was 38%-100%. The highest of similarity
coefficient (100%) was in between Bengkalis II bulk A and Pelalawan bulk A,
Pelalawan bulk B and Rokan Hulu II bulk B, Rokan Hilir bulk C and Siak bulk A.
The lowest of similarity coefficient (38%) was in between Bengkalis I bulk B and
Indragiri Hilir bulk C. In conclusion, the genetic diversity of 10 mungbean
genotypes from Riau Province was approximately 36%.
