Analisis Sekuens Barkode Dna Pada Daerah Ets (External Transcribed Spacer) Pada Tumbuhan Durik-Durik (Syzygium Sp.) Asal Riau
Durik-durik (Syzygium sp.) plant belongs to the Syzygium genus has many benefits. Syzygium is a genus that has many species making it difficult to identify using morphological characters. This study aims to analyze DNA barcode sequences in the ETS (External Transcribed Spacer) region of durik-durik plant. The research procedure begins with the provision of leaf samples, total DNA isolation, electrophoresis, PCR, sequencing and data analysis. The primers used in this study are MyrtF primers of 5'-CAT GGG CGT GTG AGT GGT GA-3' and 18SR primers of 5'-GAG CCA TTC GCA GTT TCA CAG A-3’. Data analysis used BioEdit version 7.0, BLASTn and MEGA version 6.0. The results showed that the ETS sequence has a size of 541 bp. Based on the identity value, Syzygium sp. has the closest relationship with Syzygium nervosum with an identity value of 95.57%. The species name of the durik-durik plant cannot yet be determined because there is no accession that has the identity value reaching 100%.
