Analisis Sekuens Barkode Dna Pada Daerah Gen Ndhf Pada Tumbuhan Durik-Durik (Syzygium Sp.) Asal Riau

Durik-durik plant (Syzygium sp.) is on of the important plants that live in and around Kajuik Lake located in Langgam, Pelalawan, Riau Province. In 2010, durik-durik was classified as Eugenia sp. In 2016 and 2017, the study on durik-durik were carried out through DNA barcoding analysis based on psbA-trnH intergenic spacer, matK, rbcl, ITS dan trnL-F intergenic spacer. The conclusion was that the durik-durik were members of Syzygium not Eugenia, but not yet identified to species level. Therefore, further research is carried out in this study by analysis of DNA barcode of ndhF gene to identify durik-durik from Riau. The research procedure included provision of samples, total DNA isolation, electrophoresis, PCR, sequencing and data analysis. Data was analyzed using BioEdit version 7.0, BLASTn and MEGA version 6.0. The analysis showed that the durik-durik ndhF gene had a size of 1370 bp. The alignment result indicated that the durik-durik ndhF gene sequence had highest similarity 96,63% to Syzygium forrestii. Unfortunately, there were no accessions that had 100% similarty to durik-durik. Therewere 23 different nucleotides and two of them were the important nucleotides for durik-durik identification. The species name of durik-durik are still yet not determined.
