Analisis Sekuens Barkode Dna Pada Daerah Atpb-Rbcl Intergenic Spacer Pada Tumbuhan Durik-Durik (Syzygium Sp.) Asal Riau
Durik-durik (Syzygium sp.) is a plant from the family Myrtaceae that can grow in the flooded swamp ecosystem. This plant has a good adaptability to inundation stress. Initially morphologically durik-durik has been characterized as Eugenia sp. However, after molecular identification using intergenic psbA-trnH intergenic spacer and ITS DNA barcodes, durik-durik is grouped into the genus Syzygium. This study aimed to analyze DNA barcoding sequences in the atpB-rbcL intergenic spacer region of the durik-durik plant. Leaf samples of durik-durik (Syzygium sp.) plant taken from Kajuik Lake located in Langgam Village, Pelalawan Regency, Riau Province. Samples were extracted using Genomic DNA mini kit Plant (Geneaid) then amplified using dreamTaq DNA polymerase and atpB-rbcL intergenic spacer primer pair. The PCR product was sequenced at 1st Base Malaysia through an intermediary from PT Genetika Science Indonesia, West Jakarta to determine the nucleotide sequences of durik-durik plant. The results showed that the DNA barcoding sequences of atpB-rbcL intergenic spacer region in this sample was 908 bp and could clearly separate the genus Syzygium from other genera. There is a close kinship between durik-durik (Syzygium sp.) from Riau and Syzygium forestii. However, this sequences still does not show clearly the name of the species from Syzygium sp. from Riau.
