Analisis Sekuens Barkode Dna Pada Daerah Trnq-5’rps16 Intergenic Spacer Pada Tumbuhan Pandan (Benstonea Sp.) Asal Riau

Pandan (Benstonea sp.) is one of the most important plants in Kajuik Lake located in Langgam, Riau Province, Indonesia. This plant has been previously idetified in 2017 with the result that pandan is a member of Benstonea, but has not been identified to species level. Therefore, further DNA barcoding analysis using intergenic spacer located in plant chloroplast genome need to be performed. The study was to analyze DNA barcode sequence in the trnQ-5' rps16 intergenic spacer region on the pandan plant. The stages of the research were total DNA isolation using Genomic DNA Mini Kit Plant (Geneaid), Polymerase Chain Reaction (PCR), electrophoresis, sequencing and data analysis using BioEdit version 7.0, BLASTn and MEGA version 6.0. The analysis showed that the sequence of trnQ-5 ' rps16 intergenic spacer obtained was 926 bp in size. The sequence can distinguish Benstonea genus from other genera. In this study, the species name of this pandan still cannot be determined.
